Stonewall, Britain’s most successful LGBT club who are now in their 27th season, will face Wilberforce Wanderers in the Middlesex County Division One game on November 30.
The match, the first regular non-league fixture to be played at the new Wembley, marks the FA’s link-up with LGBT charity Stonewall.
As part of the partnership, the FA will become a member of Stonewall’s Diversity Champion programme and work to create an inclusive environment for LGBT players and fans.
Former Chelsea defender Paul Elliott, head of the FA’s Inclusion Advisory Board, said: “This will be a truly iconic and memorable evening for everyone involved.
"This new relationship presents an opportunity for football to help change hearts and minds beyond the stadia, as well as ensuring that everyone who makes football happen feels included.
“The principle behind this partnership therefore embraces the philosophy that the game is for anyone, anytime, anywhere.”
While Stonewall FC manager Eric Najib said: "This event is a landmark moment for all those who have worked tremendously hard to eradicate all forms of discrimination within football, including our sponsors TeamPride [the consortium of businesses that supports Rainbow Laces.
"We are delighted that Stonewall FC will be able to showcase our talent under the stadium's famous arch.
"This game will mark the pinnacle in my 12-and-a-half years as manager of the club and I'm extremely thankful to all those involved that made this happen."
