The under-fire Portuguese boss was left fighting for his job after a humiliating 3-0 defeat at home to Tottenham.
He was forced to face a barrage of questions after the contest with reporters quizzing Mourinho on the reaction of United fans.
A large number of supporters had seen enough and headed for the exit doors as United endured their worst start to a Premier League season for 25 years.
A minority of the remaining fans who stayed behind applauded the team and the beleaguered Portuguese boss who looked emotional at the full-time whistle.
Firing back at reporters after being asked of fans leaving early he vented: "The best judge in football are the supporters, they are the best judge.
"I would do the same, losing 3-0, taking two hours from here to the centre of Manchester. I live there, I know what it is like after matches – so keep trying, you keep trying.
"We lost last season here vs Sevilla and we were booed, and we deserved it. And today the players left the pitch after losing at home and they were applauded because they deserve it.
"So you keep try, and trying, and trying."
As a journalist tried to hit back, Mourinho interrupted: "Just to finish do you know the result?
"3-0 but it also means three Premier Leagues and I won more Premier League's alone than the other 19 Premier League managers put together. Three for me and two for them"
He then stormed out of the room shouting "respect" repeatedly.
Earlier this week Mourinho ended his pre-game press conference in less than five minutes.
And he was in no mood to hang around after his worst home loss ever as a manager.
United legend Gary Neville thinks Mourinho should stay on at Old Trafford and see out his contract.
Speaking on Sky Sports, Neville said: "This is the biggest challenge Jose Mourinho has ever faced in his career.
“Ed Woodward should let him see out his contract — you can’t keep sacking managers.
“It is time to batten down the hatches. Ed and Jose need to sort it out and get the team right for the rest of the season.
“The club is bigger than any individual. It’s time for no more politics and fighting.”
